Introduction

Somatostatin Analogue
- More potent inhibitor of GH, glucagon and insulin than naturally occurring hormone

Indications Uses

1. Bleeding esophageal varices
2. Acromegaly
3. Tumors

Mechanism of action

binds Somatostatin receptors
- Inhibits secretion of gastrin, CCK, glucagon, GH, insulin, secretion, TSH, VIP
- Reduce pancreatic secretions
- Decrease GIT and gallbladder contraction
- Vasoconstriction particularly reduces portal vessel pressure

CVS - Conduction abnormalities

CNS - Mild analgesic effects (Mu)

- Abdominal cramps
- Constipation/diarrhea
- Hyperbilirubinaemia, gallstones
- Pancreatitis
- N/V

Endocrine
- Hypothyroidism
- Hyperglycaemia/hypoglycaemia
